What is Ipamorelin?

Ipamorelin is a synthetic pentapeptide and selective growth hormone secretagogue, first described by Raun and colleagues in 1998 as “the first selective growth hormone secretagogue.” It remains one of the most extensively characterised compounds of its class in the published research literature.

How does Ipamorelin work?

Ipamorelin mimics the hormone ghrelin at the growth hormone secretagogue receptor (GHSR-1a) on pituitary cells, triggering a pulse of growth hormone release. It acts on a different receptor from GHRH analogues such as CJC-1295, which is why the two are frequently studied together.

What makes Ipamorelin “selective”?

Its selectivity is its defining feature. In the foundational research, Ipamorelin released growth hormone without meaningfully raising cortisol, ACTH or prolactin — even at doses far above the level needed for GH release. Older growth hormone-releasing peptides don’t share this clean profile.

Because the two pathways are separate but converge on the same GH-secretory machinery, activating both together has been shown in preclinical models to produce greater GH pulse amplitude than either alone.
